GENETIC DIVERSITY OF ITALIAN, CANADIAN AND IRANIAN WHEAT VARIETIES USING AFLP

DNA fingerprinting, AFLP, durum and common wheat varieties, reological quality

 

AFLP techniques were used to survey genetic divergences and to determine intra specific genetic relationships within some durum wheat varieties from Italy (Mongibello, Nedda, Linea-25, Amedeo, Bronte and Ct 298), Canada (Pathfinder, Navigator, Avonlea, Wascana, Melita and Kyle), Iran (IRN 1, IRN 3,IRN 4, IRN 5, IRN 6), Durex from USA and six common wheat from Iran (IRN 2, Kenz, Opata-85, Attila-12, Pfau/sed). We employed 4 AFLP primer combinations (using the EcoRI/MseI couple restriction enzymes) on durum wheat and common wheat. A total of 434 bands were observed; 142 of these were polymorphic. Genetic similarities among all taxa ranged from 0.109 to 0.480 (between the tetraploid) and 0.129 to 0.178 (between common wheat). Phenetic trees based on genetic similarities (UPGMA, N-J) were created. Trinakria female parent of Mongibello, Linea 25 and Nedda has shown a genetic distance with them ranged between 0.225 (Linea 25) and 0.192 (Nedda). Similar behaviours was revealed within each Canadian and Iranian genotypes. Between all tested durum wheat varieties, Amedeo has shown an AFLP pattern closer to common wheat. Among durum wheat the genotypes studied, regarding the strong gluten character W (alveograph index), Mongibello, Pathfinder and Navigator have shown a significant higher difference (P£0.05 according to ANOVA mean test Student-Newman-Keuls) respectively with values of 393, 346 and 329.
