CHARACTERIZATION OF
GENE REPERTOIRES DURING DEVELOPMENT OF CITRUS FRUITS

We
carried out a random sequencing of cDNAs derived from immature and mature
citrus fruit (Citrus sinensis) to identify the gene repertoires
expressed during development. A technique of RNA fingerprinting was
optimized allowing the detection
of DNA fragments derived from RNA using cDNA synthesis and subsequent PCR
amplification. RNAs were isolated from three
cultivars with different level of pigmentation and at three developmental
stages. Thirty different primer combinations, each of the primer carrying a
selective base at 3’ end, were employed in PCR assay to amplify the
sequences generated by restriction reaction with BstYI and MseI. Sequencing
and homology search in DNA database is beginning, from which genes related to
fruit development will be identified.
