ASSESSMENT OF GENETIC DIVERSITY IN HAPLOID CLONES EXTRACTED FROM TETRAPLOID CULTIVATED POTATO

In genus Solanum cross-pollination between heterozygous tetraploid potato produce offspring that display complex segregation. Analysis of genetic variation among the haploids derived from cultivated potato (tbr) could be extremely useful to detect the allelic diversity present in them. Eight tbr varieties with wide genetic background were crossed with two accession of Solanum phureja haploid inducer to extract haploids. The populations obtained were evaluated for agronomic and resistance traits. A high variability in canopy, total yield and specificy gravity was found in each haploid population.

 

Significative difference was found in tuber yield between individuals and populations. The total yield ranged from a value minimum of 47,6 qli/ha (COI 2b) to a maximum of 753 qli/ha (V2I 39). Seven clones showed a total yield higher than c.v. Spunta (450 q/he). CFK 69.1 haploid population showed the higher medium value in total yields (563,3 qli/ha). Difference was also observed among specificy gravity of eight hapoid families. The mean of specific gravity of eight haploid populations ranged from 1,010 to 1.111. The results of evaluation for resistance to Erwinia carotovora  subsp. carotovora  (Ecc) and to PVX indicated that 28% of genotypes tested result resistant to Ecc and 24% to PVX. One haploid population was analyzed with molecular markers to examine the degree of variability present.

 

With eight AFLP combinations 257 polymorfic band were detect. Genetic similarity values were calculated and a cluster analysis was performed to generate a dendogram. The similarity index varied from 0,584 to 0,954. These haploids can be cross with complementary genotype to maximize heterozigosity or with of cultivated species to introduce desiderable traits.
